WASHINGTON — Government scientists Thursday reported two im portant advances in making a vac cine to ward off the most serious kind of malaria, still a major killer in many parts of the world. Although researchers said large- scale production of an effective vac cine may yet be years away, the Na tional Institute of Allergy and Infec tious Diseases said the new developments bring such vaccines closer to reality. One team of institute scientists, headed by Dr. Thomas McCutchan, divised a genetic engineering tech nique to cut intact genes directly out of the hereditary blueprints of the parasite that causes malaria. Another team, directed by Dr. John Dame, applied that technique to the protozoa called a sporozoite that is injected by an infected mos quito into the blood stream. Dame cloned the gene that carries instruc tions to make the protein that alerts the body’s immune system to attack the sporozoites when they enter the body. The next step, Dame said, is to produce large amounts of the pro tein and then test it to see how effec tive it will be as a vaccine to create immunity against the sporozoite. “One can’t say that this is going to be the cure-all for malaria,” Dame said in an interview. There are four species of mos- quito-born organisms that cause ma laria in humans. The current re search zeros in on the kind — plasmodium falciparum — that causes most of the malaria fatalities. Complicating the vaccine prob lem, however, is the fact that the parasites have four important stages in their life cycle and immunity against the sporozoites that start the infection in humans will not protect against the later stages. Therefore, Dame said, a vaccine against the sporozoite must be capa ble of knocking out all the parasites to protect against disease.
